1,2-ジメチルヒドラジン投与マウスにおける大腸腺腫発症に与える食餌性ナガイモの効果 OAK

ナガイモのガンに対する機能性を探索する目的で, 1,2- ジメチルヒドラジン投与マウスにおける大腸腺腫(ACF) 発症に与える食餌性ナガイモ粉末の効果を検証した.ナガ イモ粉末をAIN-93G標準飼料のコーンスターチ部分に 100%または50%置き換えて投与したところ,大腸腺腫の 発症が有意に抑制されたまた,加熱および非加熱の生ナ ガイモ粉末ともに同様の効果が認められた. DNAマイク ロアレイを用いて大腸での遺伝子の異同を調べたところ, ナガイモ投与群ではアポ卜ーシスを誘導する遺伝子群の増加が認められた.To clarify the physiological effects of dietary Chinese yam (Nagaimo) on colon cancer in vivo, the effects of dietary Nagaimo on 1,2-dimethylhydrazine (DMH)-induced aberrant crypt foci (ACF) formation in the large intestine of mice were investigated. The early molecular dialogue between soybean and the bacterium Bradyrhizobium japonicum is crucial for triggering their symbiotic interaction. Here we found a single large genomic locus that is widely separated from the symbiosis island and was conspicuously induced within minutes after the addition of genistein. This locus (named BjG30) contains genes for the multidrug efflux pump, TetR family transcriptional regulator, and polyhydroxybutyrate (PHB) metabolism. The induction of BjG30 by genistein was competitively inhibited by daidzein, although both genistein and daidzein are soybean-derived inducers of nodulation (nod) genes. Oleanolic Acid and Ursolic Acid in Commercial Dried Fruits OAK

Oleanolic acid (OA) and ursolic acid (UA) are natural triterpenoids that have preventive properties such as anti-tumor and anti-hepatitis activities. These triterpenes are known to exist in several medicinal plants and fruit skins. Dried fruits are usually eaten without removing the skin; thus, they may be good resources for oral intake of OA and UA. HPLC analysis of OA and UA contents in a variety of dried fruits showed that raisins contained higher levels of OA than the other fruits investigated, with green raisins containing the highest levels (79.0 mg/100 g). 食餌性ナガイモの大腸腺腫発症に与える効果を他のイモ類と比較しつつ,1,2-ジメチルヒドラジン投与マウスを用いて調べるとともに,ナガイモを利用した商品モデル(ナガイモ入り青汁)の効果についても検証した.その結果,ナガイモ以外のイモ類においても上記の実験動物モデルにおいて大腸腺腫抑制効果が観察されたが,腺腫数の平均はナガイモ群が一番低かった.また,商品モデルであるナガイモ入り青汁ついても腺腫抑制効果が観察された.The dietary effects of Chinese yam,nagaimo (Dioscorea opposita Thunb.),on 1,2-dimethylhydrazine (DMH)-induced aberrant crypt foci (ACF) formation in the large intestine of mice were investigated and compared with those of other potato species. Potato consumption was found to significantly suppress ACF formation in the large intestines of all mice treated with DMH. Suppression of ACF formation was most remarkable in the nagaimo group. Moreover, the commercial product (aojiru) containing nagaimo powder also suppressed ACF formation.
